Stem cell senescence refers to the irreversible growth arrest of stem cells caused by accumulated stress, DNA damage, telomere attrition, and metabolic dysfunction. Unlike healthy quiescence, senescent stem cells lose their ability to self-renew and regenerate tissues while remaining metabolically active. Senescent stem cells often secrete inflammatory factors that disrupt the stem cell niche and impair neighboring cells. With aging, increased stem cell senescence contributes to reduced tissue repair, delayed wound healing, and organ degeneration. This process affects multiple stem cell populations, including hematopoietic, muscle, and neural stem cells. Stem cell senescence is a major driver of regenerative decline and functional aging. Understanding its molecular triggers is essential for developing therapies that restore regenerative capacity and support healthy aging.
