Senescence-Associated Secretory Phenotype (SASP)

The senescence-associated secretory phenotype (SASP) refers to the bioactive molecules secreted by senescent cells, including pro-inflammatory cytokines, chemokines, growth factors, and proteases. SASP factors alter the tissue microenvironment by promoting inflammation, extracellular matrix remodeling, and disruption of normal cellular communication. While transient SASP activity can aid wound healing and immune surveillance, chronic SASP signaling contributes to age-related tissue degeneration and systemic inflammation. Persistent SASP promotes the spread of senescence to neighboring cells and accelerates aging processes. SASP is strongly associated with “inflammaging,” a hallmark of aging linked to chronic disease. Targeting SASP components through senomorphic therapies is a promising approach to reduce age-related inflammation without eliminating senescent cells entirely.
